Fluorescence Correlation Spectrometer (FCS) is a diffusion-based single-molecule technique. In this technique, spontaneous fluorescence fluctuation is collected from very few molecules as they diffuse in and out of a tiny observation volume (~1 fL) over time. The fluorescence fluctuations are correlated to obtaining dynamic information contained in the fluctuation. By analyzing the correlation curve with suitable models, one can quantitatively measure diffusion coefficient, viscosity, chemical kinetics, size, etc.
